Ezetimiba/rosuvastatina per al tractament de la hipercolesterolèmia

Abstract

La rosuvastatina (ROS) és un inhibidor competitiu i selectiu de l’hidroximetilglutaril coenzim A (HMG-CoA) reductasa, l'enzim limitant que genera el mevalonat, un precursor del colesterol. Augmenta el nombre de receptors LDL hepàtics en la superfície cel·lular per incrementar l'absorció i el catabolisme del colesterol LDL i inhibeix la síntesi hepàtica de VLDL. L’ezetimiba (EZE) actua a nivell del transportador d'esterol, el Niemann-Pick C1-Like 1 (NPC1L1), responsable de la captació intestinal de colesterol i fitosterols de manera que inhibeix selectivament l'absorció intestinal del colesterol i els esterols vegetals relacionats. La combinació d’ambdós fàrmacs amb mecanismes diferents produeix una reducció complementària del colesterol.
